How do you set up an eraser so that whatever the cursor pixels touch is completely removed in one click? I'm constantly having to feather things out to alpha with several clicks...like reverse anti-aliasing.

In the Tool Options:
1) Select a hard brush (eg round brush hardness 1.00)
2) There is a an option Hard Edge. Tick that.

I would say,

Use a suitable brush, one that does not have any anti-aliasing and just to make sure, enable Hard edge
